The oxysterol receptor GPR183 in inflammatory bowel diseases

چکیده
Immune cell trafficking is an important mechanism for the pathogenesis of inflammatory bowel disease (IBD). The oxysterol receptor GPR183 and its ligands, dihydroxylated oxysterols, can mediate positioning immune cells including innate lymphoid cells. has been mapped to IBD risk locus, however another gene, Ubac2 encoded on reverse strand associated with Behçet's disease, therefore role as a genetic factor requires validation. production ligands are up-regulated in human murine colitis. Gpr183 inactivation reduced severity colitis group 3 cells-dependent IL-10 but not dextran sodium sulphate Irrespectively, knockout strongly accumulation intestinal tissue health all models. In conclusion, genetic, translational experimental studies implicate GPR183-dependent migration might be therapeutic drug target IBD. The Bowel Microbiota and Inflammatory Bowel Diseases
The human bowel contains a large and biodiverse bacterial community known as the microbiota or microbiome. It seems likely that the microbiota, fractions of the microbiota, or specific species comprising the microbiota provide the antigenic fuel that drives the chronic immune inflammation of the bowel mucosa that is characteristic of Crohn's disease and ulcerative colitis. At least twenty years...

Uncontrolled activation of mucosal effector cells has been identified as the main pathogenic mechanism involved in the initiation and perpetuation of mucosal inflammation in inflammatory bowel diseases (IBD). The sustained activation of these cells leads to the aberrant production of various pro-inflammatory mediators, which co-ordinated action amplifies the inflammatory process. In this settin...
